Sandwich TR-FRET immunoassay designed for the semi-quantitative detection of Phospho-RIPK1 (S166) in cell lysates. The assay uses the advanced THUNDER® TR-FRET technology and a matched antibody pair specifically optimized and validated for TR-FRET. Full assay validation can be found in the Technical Data Sheet. Detailed assay protocols are described in the Generic User Manual. The assay is compatible with both adherent and suspension cells, and can be run under either a one-plate or two-plate protocol. Both methods are “no-wash”.

Inhibition of Phospho-RIPK1 (S166) in HT-29 cells
Data show that treatment of HT-29 cells with Necrostatin-1S inhibits phosphorylation of RIPK1 at S166 by TNF-alpha. in the presence of SM-164 and Z-VAD.
Z'-factor determination in HT-29 cells
The Z’-factor value of 0.74 indicates that the assay is robust and suitable for HTS.
HT-29 control lysate titration (QC Test)
Data show a linear relationship between lysate dilutions and TR-FRET ratio values. following 1/2 dilution.
